Purpose
The type 1 cannabinoid receptor (CB1R) is the receptor that is activated by .9-tetrahydrocannabinol (THC), the chemical in Cannabis that produces the 'high' Cannabis users experience. THC, and the related cannabidiol (CBD) are known as a cannabinoids and at least 120 other cannabinoids have been identified in the Cannabis plant. Cannabinoids are being explored for their potential use to treat epilepsy, pain, anxiety, and numerous other ailments. Unfortunately, our knowledge of how cannabinoids work at CB1R is limited. In this project, we will use computer-assisted drug design programs and drug discovery experiments to understand how cannabinoids from the Cannabis plant work at the molecular level. Mutant forms of the receptor will be used to help us understand which parts of CB1R are required for cannabinoids to produce cellular effects. From this study we will gain critical knowledge about how cannabinoids work with CB1R. This knowledge will lead to improved understanding of cannabinoid biology, safe and effective use of cannabinoids, and the potential for new cannabinoid drugs.
